Conformation Specific Antibodies

Early research yielded a "first generation" of antibodies which could not specifically recognize proteins based on activation state or conformation. "Second generation" antibodies were able to recognize activated proteins, but only by virtue of their phosphorylation status. Although a great improvement, the application of phosphoantibodies is limited in scope. Unfortunately, many known cancer targets or disease-causing proteins are not activated by phosphorylation, but by the binding of a receptor-specific ligand, which alters the three-dimensional structure or conformation of the molecule. Until now, there has been no way to systematically generate antibodies which can conformationally distinguish signaling proteins in their activated or inactivated state.
